2. Modification of H3-R4

 

2.1 H4R3me2a

It is asymmetrically methylated by PRMT1. H4R3me2 acts as a transcription activating mark of several ER regulated genes and is essential in vivo for the establishment and maintenance of open chromatin domains marked by H3 and H4 acetylation. (1)

2.2 H4R3me2s

H4R3me2s serves as a direct binding target for the DNA methyltransferase DNMT3A, which interacts through the ADD domain containing the PHD motif. Loss of the H4R3me2s mark through short hairpin RNA–mediated knockdown of PRMT5 leads to reduced DNMT3A binding, loss of DNA methylation and gene activation. In primary erythroid progenitors from adult bone marrow, H4R3me2s marks the inactive methylated globin genes coincident with localization of PRMT5. (2)

3. Interplay between H3-R4 and other proteins:

 

3.1 DNMT3A: as both a reader and a writer of repressive epigenetic marks, thereby directly linking histone and DNA methylation in gene silencing. (3)

3.2 SKB1: Arabidopsis Shk1 binding protein 1 (SKB1) catalyzes histone H4R3 symmetric dimethylation (H4R3sme2). SKB1 promotes flowering by suppressing FLC transcription. SKB1 binds to the FLC promoter, and disruption of SKB1 results in reduced H4R3sme2, especially in the promoter of FLC chromatin. Thus, SKB1 mediated H4R3sme2 is a novel histone mark required for repression of FLC expression and flowering time control. (4)

3.3 PRMT7 transcriptional regulation of imprinted genes: Chromatin immunoprecipitation (ChIP) revealed that PRMT7 and dimethylated H2AR3 and H4R3 are enriched at target DNA repair genes in parental cells, whereas PRMT7 knockdown caused a significant decrease in PRMT7 recruitment and H2AR3/H4R3 methylation.
