OBJECTIVE: To investigate the expression of high mobility group box-1 (HMGB1) mRNA in liver tissue of rats with sepsis induced by Janus kinase / signal transduction and transcription activator (JAK / STAT) pathway in patients with cecal ligation (CLP) Acute liver damage. Methods: The rats were randomly divided into normal control group, CL P sepsis group, JAK2 kinase inhibitor AG490 and STAT inhibitor rapamycin (RPM) treated group. The liver HMGB1 mRNA was determined by reverse transcription polymerase chain reaction (RT-PCR), and the liver function was measured by automatic biochemical analyzer. Results: Compared with the normal control group, the expression of HMGB1 mRNA was significantly increased at 6 ~ 48h after CL P (P <0.01). Serum aspartate aminotransferase (AST) increased significantly at 6 ~ 48h (P <0.01). 0), alanine aminotransferase (AL T) and AST were significantly increased at 24 h (P <0.01). Compared with CL P group, the levels of HMGB1m RNA and ALT were significantly decreased in AG490 pretreatment group (P <0.01), and the AST was significantly lower at 24 h and 48 h (P <0.01) The expression of HMGB1m RNA was significantly inhibited at 6 h and 24 h after RPM intervention (P <0.05 and P <0.01). ALT and AST decreased at 24 h and 48 h (P <0.01) 1 and P <0 05). Conclusion: Inhibition of JAK / STAT pathway activation can downregulate the expression of HMGB1 mRNA in liver tissue and contribute to the alleviation of acute liver injury induced by CL P.
