Protein function

May play a role in intercellular signaling and in connecting cells with the extracellular matrix. May take part in the regulation of cell motility, growth and differentiation. Binds hyaluronic acid.

Protein tissue specificity

Cerebral white matter and plasma. Isoform V0 and isoform V1 are expressed in normal brain, gliomas, medulloblastomas, schwannomas, neurofibromas, and meningiomas. Isoform V2 is restricted to normal brain and gliomas. Isoform V3 is found in all these tissues except medulloblastomas.

Involvement in disease

Wagner vitreoretinopathy (WGVRP) [MIM:143200]: A rare vitreoretinopathy characterized by an optically empty vitreous cavity with fibrillary condensations and a preretinal avascular membrane. Other optical features include progressive chorioretinal atrophy, perivascular sheating, subcapsular cataract and myopia. . Note: The disease is caused by mutations affecting the gene represented in this entry. The pathological mechanism involves a quantitave imbalance of the normally occurring splice variants (PubMed:22739342). .

Protein sequence and domain

Belongs to the aggrecan/versican proteoglycan family. / Contains 1 C-type lectin domain. / Contains 2 EGF-like domains. / Contains 1 Ig-like V-type (immunoglobulin-like) domain. / Contains 2 Link domains. / Contains 1 Sushi (CCP/SCR) domain.

Protein post-translational modifications

Phosphorylated by FAM20C in the extracellular medium.

Result figure:
Versican.pdf

Figure description:

Versican is present within the cytoplasm of embryonic heart cells. This pattern may be real, as another extracellular matrix factor, Fibronectin, also has strong intracellular accumulation. However, more tests are necessary to determine if the intracellular Vcan signal is real, including staining in Vcan knockdown or knockout organisms, Western blot, and immunostaining at other developmental stages.
GFP marks migrating endocardial cells, DAPI marks nuclei.
