Applications
| GenScript Goat Anti-Rabbit IgG (H&L) [HRP] is suitable for immunoblotting (western and dot blots), ELISA, immunoperoxidase electron microscopy, immunohistochemistry, and other peroxidase-antibody based enzymatic assays requiring lot-to-lot consistency. The investigator must determine the ideal working concentration for each specific application. The ideal working concentration must take into account such factors as secondary antibody affinity, antigen concentration, sensitivity of the detection method, temperature, and the length of the incubations. The suitability of this antibody for applications other than those listed below has not been determined. The following concentration ranges are recommended starting points for this product. C2C12 mouse muscle cells were transfected with a plasmid expressing connexin 43 IRES FusionRed under the control of a mature muscle promoter MCK. Normally C2C12 cells do not express Connexin 43 in mature myotubes. They do express connexin 43 in immature myoblasts. The negative control are the same cells with the FusionRed tag only. There is still some staining of connexin 43, but not in the mature myotubes.

The sample was C2C12 mouse muscle cells transfected with a plasmid that expresses connexin 40 IRES FusionRed under a mature muscle promoter MCK. The negative control were the same cells with just the FusionRed tag. It appears that there is light non-specific binding.

Human multiple myeloma cell line KMS11 was cultured in RPMI-8226 media and then lysates are made in RIPA buffer supplemented with phosphatase and proteinase inhibitors. 2ug of protein was loaded in two wells and electrophoresed for 30min at 200V BIORAD electrophoresis unit. I used a dilution of 1:1000, incubating the membrane with the antibody overnight at 4 degrees.
I detected along with my specific protein band, also one unspecific which maybe adjusting the condtions I could get rid of it.
